1、中南大学博士学位论文DIXDC1与DISC1在匹罗卡品致痫小鼠模型海马结构中的动态表达变化姓名:向田申请学位级别:博士专业:神经病学指导教师:肖波201205』∑生剑堂僮途塞虫塞擅噩摘要目的研究DIXDC1(Dishevelled—Axindomain—containing1)与DISCl(Disrupted—in-Schizophrenia1)在匹罗卡品致痫小鼠模型海马结构中的动态表达改变,探讨其在颞叶癫痫发病中的作用。方法制备匹罗卡品致痫C57BL/6成年小鼠模型67只,对照组小鼠56只,取其脑海马
2、结构为研究对象。采用免疫组化观察DIXDCl在癫痫模型小鼠SE后3天、7天、14天及28天海马结构中的表达分布、通过Westemblot、Real.timePCR方法半定量检测DIXDC1、DISCl蛋白和Dixdcl、Mcm2mRNA在海马结构中各时间点的表达变化。通过SPSSl6.0软件对癫痫模型小鼠海马结构中DIXDCl和DISCl的蛋白表达改变、Dixdcl和Mcm2mRNA的转录水平进行相关性分析。结果1.免疫组化显示DIXDCl表达于海马齿状回颗粒细胞层神经元胞浆内。癫痫模型小鼠SE后各时间
