1、2006级硕上研究生学位论文罗格列酮钠对实验性l型糖尿病大鼠胰岛13细胞凋广的影响罗格列酮钠对实验性1型糖尿病大鼠胰岛13细胞凋亡的影响摘要目的:观察罗格列酮钠(rosiglitazonenatrium,RSG)对实验性1型糖尿病(type1diabetesmellitus,T1DM)大鼠胰岛13细胞凋亡的影响。方法:6周龄SD大鼠72只随机分成3组,正常对照组(A组)24只,糖尿病对照组(B组)24只和RSG治疗组(C组)24只。B、C组经腹腔注射2%链脲佐菌素(streptozotocin,STZ)溶液65mg/kg,建立1型糖尿病模型。FBG≥
2、16.7mmol/L为造模成功。C组成模当天开始给予RSG7mg/kg.d1)灌胃治疗,A、B组给予相应剂量的溶剂灌胃。注射STZ后3天及用RSG治疗后2周、5周、7周、10周和13周,检测大鼠体重(Weight,Wt)、空腹血糖(fastingbloodglucose,FBG)和空腹胰岛素(fastinginsulin,FINS),A、B、C三组各随机处死动物4只。留取胰腺标本行病理切片和超薄切片。光镜下观察胰岛病理形态学改变,透射电镜观察胰岛13细胞超微病变,用TUNEL方法检测胰岛13细胞的凋亡,用免疫组化法检测胰岛中FINS、Fasl的表达,
3、并进行量化分析。结果:与B组相比,C组经RSG干预2周后,①糖尿病大鼠的饮水量逐渐减少,体重逐渐增加(PO.05);④胰岛Fasl表达逐渐减少,Fasl阳性面积密度与相对灰度值均降低,经简单相关性分析,前者与胰岛素面积密度呈负罗格列酮钠对实验性1
