1、摘要目的:建立大鼠离体肝脏保存模型,探讨丹参和Ho_1的mRNA及其蛋白在离体肝脏保存时的关系,进一步明确丹参对肝脏的保护作用的生物机制,为进一步提出丹参在缺血再灌注对肝脏保护作用提供理论依据。方法:一、通过逆转录一聚合酶反应(RT-PCR)测定各组时间段保存后HO-lmRNA表达情况;二、通过蛋白免疫组化监测各组时间段保存后Ho_1蛋白表达水平;三、用光镜观察各组不同时相肝细胞、肝窦皮细胞的形态变化;四、Ho_1的mRNA表达相对值及其蛋白的表达灰度值与肝脏损伤评分相关分析。结果:一、成功建立大鼠
2、离体肝脏保存模型;二、RT—PCR结果显示,丹参使HO-lmRNA的表达上调,经ZnPP预处理组微量表达;三、免疫组化结果显示丹参使Ho_1蛋白相对应于HO-lmRNA的表达上调而上调;四、病理切片可以证明经丹参处理后肝标本比对照组的干细胞、肝窦、损害明显较小,而经ZnPP预处理组较其他两组损害严重;五、在不同组相应的时间段HO-lmRNA的表达与肝组织损伤评分呈负相关,Ho_l的蛋白质表达灰度值与肝组织损伤呈正相关。结论:丹参可以诱导Ho_1的mRNA和蛋白的过表达,这种表达与肝组织损伤的关系,可
