1、中文摘要持续低流量灌注对离体鼠心保存效果的实验研究摘要目的:单纯低温静置保存是效果已被验证的心脏保存方法,但是术后早期的心功能抑制还是比较常见的,而且此方法的心脏保存时间较短,只有4.6h。持续低流量灌注是新型的心脏保存方法,本课题就是采用改良Langendorff装置,以HTK液和FWM液为心脏保存液,比较单纯低温静置保存和持续低流量灌注两种不同的保存方法对离体鼠心的保存效果,并对HTK液和FWM液的心肌保存效果进行评价。方法:32只SD雄性大鼠随机分成4组(每组8只),HTK液单纯低温静置组(H1组)
2、,HTK液持续低流量灌注组(H2组),FWM液单纯静置组(F1组),FWM液持续低流量灌注组(F2组)。开胸取出心脏后迅速转移并固定于Langendorff灌流装置上,用K.H液经主动脉恒压灌注30min,再经主动脉根部灌注4。C保存液,F1、F2组灌注FWM液,H1、H2组灌注HTK液,待心脏停跳后,在4℃条件下,F1,H1组心脏分别浸于FWM液和HTK液中,静置保存8h;F2,H2组心脏以lOcmH20的压力分别持续灌注FWM液和HTK液,灌注8h。8h后,重新将鼠心移至Langendorff装置上,
3、复灌K.H液60min。于灌停前及复灌后测定心率(HR)、冠脉流量(CF)、左心室发展压(LVDP)、心率室内压乘积(RPP)、左心室内压最大上升速率(+dp/dtm。x)、左心室内压最大下降速率(.dp/dtm弧)等指标,并计算各种参数的恢复率,同时留取冠脉流出液测定心肌酶(CK.MB,LDH)漏中文摘要出量。灌注结束后,取心肌组织测定含水量并制作光镜标本观察心肌组织结构变化。结果:复灌后,Fl组8只心脏均未复跳,心功能恢复率为O。复灌过程中我们发现,心脏颜色苍白,触之质硬,如“石样心"。心肌含水量,心
