1、摘要本论文采用多种高尿酸血症模型、急性痛风性关节炎模型、炎症模型和疼痛模型,结合现代医学多种手段,研究加味四妙散抗痛风的活性,考察加味四妙散对痛风的影响及其作用机理。采用脲酸酶抑制剂和酵母膏灌胃造成小鼠高尿酸血症,采用腺嘌呤+乙胺丁醇灌胃造成正常大鼠高尿酸血症模型。各种不同机制的高尿酸血症实验均表明,加味四妙散能明显降低血尿酸,使血清尿酸水平下降,同时还能抑制黄嘌呤氧化酶水平。按照McCartyDJ造模方法,给家兔膝关节注射尿酸盐结晶,建立急性痛风性膝关节炎家兔模型,造模2h后收集关节液,测其中IL.1p、IL.8和TNF.a的含量。实验结果表明,加味四妙散能
2、明显降低降低炎性因子的含量,缓解炎性症状;将大鼠右后足皮下注射尿酸盐结晶后诱导的大鼠急性足肿胀模型,加味四妙散能明显抑制大鼠足肿胀度,阻止炎症状态进一步发展。利用醋酸扭体化学刺激疼痛模型和甩尾法热刺激疼痛模型,实验表明,加味四妙散能明显抑制醋酸扭体反应次数,显著延长热刺激潜伏期,表明加味四妙散有较好的镇痛作用。加味四妙散对实验性痛风具有较好的治疗作用,其作用机制主要是与降低血尿酸、抑制黄嘌呤氧化酶活性、降低急性痛风性关节炎中炎性因子的含量等有关,并且加味四妙散有良好的抗炎和镇痛活性,对痛风的治疗有利。关键词:加昧四妙散,痛风,造模,高尿酸血症,炎症Abstra
