1、万方数据大连医科大学硕士学位论文脑组织中SOD、CAT、MDA、T.AOC、8-OHdG的含量进行测定。(4)应用实时荧光定量PCR、Westemblotting、免疫组织化学技术在基因和蛋白质水平上检测p53和bcl.2基因的表达,进一步解释LEO对于B[a]P染毒小鼠脑损伤的保护机制。结果:(1)本文建立了较为可靠的小鼠苯并芘亚慢性暴露模型,并应用LEO对B[a]P所致的小鼠脑组织损伤进行了有效的化学预防干预。(2)应用了高效液相色谱荧光内标法检测小鼠脑组织中3-OHB[a]P和(+).anti.BPDE.DNA的含量。与正常组相比,B
2、[alP单独染毒组小鼠脑组织中的3-OHB[a]P和(+)-anti.BPDE.DNA的含量增加(尸<0.05);与B[a]P单独染毒组相比,三组LEO保护组中3-OHB[alP的含量逐渐增加(尸<0.05),而(+).anti.BPDE.DNA的含量逐渐减少(尸<0.05)。(3)氧化应激酶类实验:与正常组相比,B[a]P染毒组可引起小鼠脑组织中的SOD、CAT和T.AOC减少(尸<0.05),而MDA、8.0HdG含量增加(尸<0.05);与B[a]P染毒组相比,三组LEO保护组中的SOD、CAT和T.AOC增加(P<0.05),而MD
