1、转录因子Tbx18在小鼠胚胎发育过程中的时空表达蒲荻1,杜建霖2,李晓群3,翁敏杰4,佘强(400010重庆,重庆医科大学附属第二医院心血管内科)[摘要]目的检测胚胎期小鼠Tbx18在mRNA及蛋白水平的表达情况,了解Tbx18在小鼠胚胎发育过程中的时空表达模式。方法用整体原位杂交技术检测小鼠整胚TBX18mRNA的表达,用X-gal染色检测Tbx18-Cre/Rosa26R/LacZ双杂基因谱系示踪模型小鼠胚胎的报告基因LACZ蛋白的表达,用荧光定量PCR检测小鼠胚胎期心脏mRNA的定量表达。结果发现小鼠胚胎转录因子Tbx18在9.5~1
2、0.5d最早主要在前体心外膜表达,由前体心外膜逐渐迁移定位于心外膜;在11.5~12.5d小鼠胚胎Tbx18主要定位于心脏、上下肢、体节及上下唇鄂区域;大于12.5dTbx18除了上述部位外还定位于肾、输尿管、膀胱等区域。结论转录因子Tbx18在小鼠胚胎发育过程中的表达具有时空特异性,对小鼠心脏的发育可能起着至关重要的作用;同时在上下肢、体节、唇鄂、肾、输尿管、膀胱的发育中也可能起非常重要的作用。[关键词]转录因子Tbx18;整体原位杂交;小鼠;胚胎发育;时空表达[中图法分类号]【文献标志码】ASpatiotemporalExpressio
