1、山羊IGFBP-6基因的克隆及其组织表达的发育性变化动物科学(特种经济动物养殖方向)2008级游杰指导教师张红平教授摘要:IGFBPs在体内循环系统中能与IGFs结合,延长IGFs的半衰期,调节IGFs的代射清除率,提高IGFs的生物学效应。肝脏和局部组织分泌的IGFBPs共同调控各个组织的生长与发育,从而促进动物机体的生长。本研究以培育品种南江黄羊为试验材料,应用RT-PCR和克隆技术克隆山羊IGFBP-6基因的全编码区序列,采荧光定量PCR技术研究该基因在南江黄羊生后5个时间点(0d、30d、60d、90d、120d)和6个组织(肝脏、肺、心脏、背最长肌
2、、半膜肌和臂三头肌)的发育性变化。结果表明:克隆得到南江黄羊IGFBP-6基因的CDS全序列长度为711bp,编码235个氨基酸,该基因的核苷酸、氨基酸序列同绵羊的一致性为97%,与其他物种间的一致性也较高;在各个时间点的各个组织中都检测到IGFBP-6基因的表达,但相对表达量存在一定的差异。IGFBP-6在背最长肌的基因表达量远远高于肝脏、心脏、脾脏、肺脏和其它肌肉等组织的表达量(0.01

6、ung,heart,longissimusdorsi,semimembranosus,musculustriceps)andatdifferenttimepoints(0d、30d、60d、90d、120dafterbirth).Theresultsindicate:(1)WehaveclonedtheCDSfullsequenceofIGFBP-6(711bp),whichencodingapeptideconsistsof235aminoacidresidues.ThenucleotideandaminoacidsequenceofIGFBP-6poss
