2、nming650201,ChinaAbstract:Chitinases(EC3.2.1.14),foundinawiderangeoforganisms,catalyzethehydrolysisofchitinandplayamajorroleindefensemechanismsagainstfungalpathogens.Thealignmentandtypicaldomainswereanalyzedusingbasiclocalalignmentsearchtool(BLAST)andsimplemodulararchite
3、ctureresearchtool(SMART),respectively.Onthebasisoftheannotationsofrice(OryzasativaL.)andArabidopsisgenomicsequencesandusingthebio-softwareSignalP3.0,TMHMM2.0,TargetP1.1,andbig-PiPredic-tor,25outof37and16outof24openreadingframes(ORFs)withchitinaseactivityfromriceandArabid
4、opsis,respectively,werepredictedtohavesignalpeptides(SPs),whichhaveanaverageof24.8aminoacidsattheN-terminalregion.Someofthechitinasesweresecretedextracellularly,whereassomewerelocatedinthevacuole.Thephylogenicrelationshipwasanalyzedwith61ORFsand25knownchitinasesandtheywe
5、reclassifiedinto6clustersusingClustalXandMEGA3.1.Thisclassificationisnotcom-pletelyconsistentwhencomparedwiththetraditionalsystemthatclassifiesthechitinasesinto7classes.Thefrequencyofdistribu-tionofaminoacidresidueswasdistinctindifferentclusters.Thecontentsofalanine,glyc
6、ine,serine,andleucinewereveryhighineachcluster,whereasthecontentsofmethionine,histidine,tryptophan,andcysteinewerelowerthan20%.Eachclusterhaddistinctaminoacidcharacteristics.Alanine,valine,leucine,cysteine,serine,andlysinewererichinClustersⅠtoⅥ,respectively.Keywords:Chit
