1、灵芝三萜类化合物对AD模型大鼠脑组织中CAT,ACHE,SOD,MDA的影响【摘要】 目的:探讨灵芝三萜类化合物(GLT)对阿尔茨海默病(AD)的防治作用,为临床新药的开发和应用提供理论依据。方法:将经Morris水迷宫筛选合格的SD大鼠80只分为8组,正常对照组、假手术组,模型组、GLT低、中、高(GLT0.25、0.5、1.0g/kg)剂量组、阳性药组(健脑胶囊组0.54g/kg)和溶媒组(食用油10ml/kg),除正常对照组和假手术组外,其余各组大鼠经侧脑室注射Aβ25-352μl制备AD大鼠模型,正常对照组侧脑室注射等量生理盐水,术后次日开始
2、灌胃给药。每天1次,连续给药20d后,处死动物取脑组织制备匀浆。采用光化学法检测脑匀浆中过氧化氢酶(CAT)、乙酰胆碱酯酶(ACHE)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果:与正常对照组比较,模型组脑组织中CAT活性降低,ACHE活性升高,SOD活性降低,MDA含量升高。与模型组比较,三个GLT给药组脑组织中,CAT活力均升高,ACHE活力均下降,SOD活性均升高,MDA含量均降低。结论:灵芝三萜类化合物对AD模型大鼠具有增强清除自由基的能力,抗脂质过氧化及促进胆碱能神经系统功能的恢复。【关键词】阿尔茨海默病;灵芝属;脑;过氧化氢酶

5、MD,received0.54g/kgofJiannaocapsules),andsolutemediatedgroup(groupSM,received10ml/kgofedibleoil).AlloftheSDratsexceptthoseingroupNCandgroupSOweremicroinjectedwithbeta-amyloidpeptidesegment25-35(Aβ25-35)intolateralventricletoproduceADmodel,andinstead,ratsingroupNCwereinjectedwith
