1、三羟异黄酮对高转移卵巢癌细胞株HO8910PM的侵袭、转移及MTA1、nm23H1基因表达的影响【摘要】目的:研究三羟异黄酮(genistein)对人高转移卵巢癌细胞HO8910PM转移相关能力的影响及其作用的机制。方法:以台盼蓝拒染法检测genistein对高转移卵巢癌细胞HO8910PM细胞增殖的影响;用Transwell小室法检测genistein对HO8910PM细胞的侵袭能力和趋化运动能力的影响;RTPCR分析MTA1及nm23H1mRNA的表达。结果:50μmol/L的genistein作用细胞6h后能抑制HO8910PM细胞体外
2、侵袭人工基底膜和趋化运动能力,抑制率分别为(21.08±3.12)%和(17.15±2.01)%;25~100μmol/Lgenistein作用HO8910PM细胞24h后,明显下调MTA1mRNA的表达,上调nm23H1mRNA表达水平。结论:genistein能抑制高转移卵巢癌细胞HO8910PM的侵袭和运动能力。genistein抗肿瘤侵袭转移的作用机制与MTA1mRNA的表达下调和nm23H1mRNA表达上调有关。【关键词】异黄酮;侵袭;运动;转移;卵巢癌;MTA1;nm23H1Effectsofgenisteinontheabilities
