2、r groups treated with Yinlongxian capsules at different dosages (1.2,0.6 and 0.3 g/kg respectively)for 6 months and the rats in each group were observed for two more weeks after the stop of the medication.During the experimental period,the general status was mornitore
3、d and the body weight of the rats was measured each weekend. The blood cells and various biochemical indexes of the blood and the histological changes in 14 major organs of the rats were determined and observed, respectively.Results: In the groups treated with Yinlongx
4、ian capsules,the growth rate and liver indexes of the rats were slightly decreased compared with that of controlgroup.No significant changes in the other blood or organ indexes were founded. Additionally, mild diarrhea was obsereved in some of the rats in the group tre
5、ated with high dosage of Yinlongxian capsule.Conclusions:Yinlongxian capsule should be safe in its clinical use at the recommended dosage. Key wordsYinlongxian capsule;longterm toxicity;rat 银龙仙胶囊的成分是银杏叶提取物、霪羊霍和冰片,由沈阳麦迪医药公司研制,主要用于心血管系统疾病的预防和治疗。目前已经完成了临床前疗效研究资料。为了进一步
6、开展该胶囊的临床试验研究,我们首先进行了小鼠口服银龙仙胶囊的毒性实验,结果表明该药以70 kg的成人推荐剂量的80倍灌胃给药30天,小白鼠体重明显减轻(可达15%),且部分小白鼠有腹泻。以70 kg的成人推荐剂量的40倍灌胃给药30天,小白鼠体重减轻不显著,也没有观察到其它不良反应。在此基础上,我们于2006年8月至2007年4月,参考有关文献报道,进行了银龙仙胶囊的长期毒性研究,目的是观察银龙仙胶囊长期灌胃给药是否会有由于药物蓄积而对机体产生毒性反应及其严重程度,确认毒性反应的靶器官及其损害的可逆性,确定无毒性反应剂量,为临床研究
7、提供依据〔1~3〕。 1材料与方法 1.1材料 1.1.1药物 银龙仙胶囊,由沈阳麦迪医药公司提供,批号2006051、2006052、2006053,临用时用淀粉糊混悬至0.24 g/ml、0.12 g/ml和0.06g/ml的浓度。 1.1.2动物 Wistar大白鼠(80只动物合格证:SCSK,辽2003-0016),8周龄,体重70~80 g,雌雄各半,由沈阳医学院实验动物中心提供。实验条件:雌雄分笼饲养,每笼5只。室温:(25±2)℃,相对湿度:(65±5)%,标准饲料,全营养喂养,自由饮水,笼具每周清洗消毒1
8、次,紫外灯灭菌10 min。通风良好,室内配有空调。实验前动物在实验室饲养房内观察7天,无异常者用于本试验。 1.1.3仪器 FA1004型上皿电子天平,上海精科天平厂生产;光镜、普通切片机由中国医科大学第二附属医院病理科提供。
