2、1)表达的作用.方法采用脂质体法转染SGC7901细胞,并筛选阳性克隆,Westernblot检测survivin蛋白的表达,RT-PCR检测MDR-1mRNA的表达,透射电镜观察转染后对SGC7901细胞的凋亡诱导作用,MTT法检测转染细胞对泰索帝的敏感性.结果survivin蛋白的表达在重组质粒组比空质粒组和空白对照组明显降低(P<O.05),重组质粒组MDR一1mRNA表达也较其他2组明显降低(P<0.01);透射电镜下可见重组质粒组细胞呈凋亡晚期变化,其MDR指数为0.196±0.013,明显低于空质粒组的2.958士0.0
3、24和空白对照组的3.126±0.019(P<O.01);重组质粒组对泰索帝的ICs.值为(16.7±1.98)ng/ml,也明显低于空质粒组的(49.9士1.21)ng/ml和空白对照组的(55.7±1.89)ng/ml(P<0.01).结论survivin反义核酸可诱导SGC7901细胞凋亡,增强泰索帝化疗敏感性,可能逆转耐药.【关键词】生存素反义凋亡泰索帝多药耐药基因【中图分类号】R735.2【文献标识码】AEffectsofSurvivinAntisenseRNAonApoptosisandReversingDrugResi
