2、elyisolatedfromthestromalvascularfraction(SVF)ofhomogenizedadiposetissue.FreshlyisolatedADSCdisplaysurfacemarkersthatdifferfromthoseofculturedADSC,butbothcellpreparationsarecapableofmultipotentialdifferentiation.Recentstudieshaveinferredthattheseprogenitorsmayresideinaperivascularlocationwher

3、etheyappearedtocoexpressCD34andsmoothmuscleactin(α-SMA)butnotCD31.However,thesestudiesprovidedonlylimitedhistologicalevidencetosup-portsuchassertions.Inthepresentstudy,weemployedimmunohistochemistryandimmunofluorescencetodefinemorepreciselythelocationofADSCwithinhumanadiposetissue.Ourresultss

4、howthatα-SMAandCD31localizedwithinsmoothmuscleandendothelialcells,respectively,inallbloodvesselsexamined.CD34localizedtoboththeintima(endothelium)andadventitianeitherofwhichexpressedα-SMA.ThenichemarkerWnt5awasconfinedexclusivelytothevascularwallwithinmuralsmoothmusclecells.Surprisingly,thewi

5、delyacceptedmesenchymalstemcellmarkerSTRO-1wasexpressedexclusivelyintheendotheliumofcapillariesandarteriolesbutnotintheendotheliumofarteries.TheembryonicstemcellmarkerSSEA1localizedtoapericyticlocationincapillariesandincertainsmoothmusclecellsofarterioles.Cellsexpressingtheembryonicstemcellma

6、rkerstelomeraseandOCT4wererareandobservedonlyincapillaries.Basedonthesefindingsandevidencegatheredfromtheexistingliterature,weproposethatADSCarevascularprecursor(stem)cellsatvariousstagesofdifferentiation.Intheirnativetissue,ADSCatearlystagesofdifferentiationcandifferentiateintotissue-specifi
