2、NIMMUNOASSAYSABSTRACTQuantumdots(QDs),asaclassofnanomaterialswithmanyuniqueproperties,havebeenexploredpotentialapplicationsinbiomoleculardetection.Incompassionwithorganicdyes,QDsoffersuchadvantagesasbetterfluorescencestabilities,narrowemissionbandwidthsandbroadabsorptionspectra.Asa
3、markerappliedinimmunediagnoseandimmunoassay,itisatrendthatQDsareinsteadofthreetraditionallabeledtechnologies.ItisimportanttoresearchthequantificationalimmunedetectionbyQDs.HereinnovelimmunoassayswereestablishedbyQDsfortheirfluorescenceproperties.InfluencesofpH,buffers,temperature,s
4、avetime,aminoacidsandproteinsonfluorescenceofQDswerestudied.QDslinkedwithproteinsbyEDC.CompetitiveinhibitionELISAmethodofmagneticparticles-SAandCdTe-SAtoBiotin-HRPanddotimmuno-percolationwereusedtodetectthebiologicactivityofproteinsafterlabeling.TheresultsprovedthattheCdTe-ptoteinc
5、ompositecouldbeappliedinimmunoassaysasithadgooddispersivityandgoodbioactivity.UtilizingthefluorescencepropertiesofQDs,twonovelimmunoassayswereestablished,whichweretheaggregation-basedimmunoassayandfluorescencequenchingimmunoassay.ThesetwomethodswereusedtodetectImmunoglobulin(IgG)an
6、dHBsAg.Theaggregation-basedimmunoassayfordetectingMouseIgGusingQDshasbeendevelopedbyusingtwomatchedantibodies.TheGoatanti-MouseIgG(GMIgG)labelledquantumdotswasmixedwithMIgG.AftertheimmunoreactionbetweenGMIgGandMIgG,thequantumdotsaggregatedtoformanetwork,whichcouldquenchthefluoresce
7、nceofthemselvesinthereactionsystem.ThemoreMIgGwas,thebiggerthenetworkwas,andthefluorescenceintensityofquantumdotsdecreasedlinearly.TheconcentrationofMIgGcouldbedetectedbythemeasurementofQDsfluorescenceintensityat567nm,andthelimitdetectingconcentrationwas1.3nmol/L.QDs-GNPsfluorescen
