2、应组,用免疫荧光和激光共聚焦显微镜等技术测定皮层顶叶Bcl-2和Caspase-3表达荧光强度和阳性细胞计数。结果显示,低氧组和低氧预适应组Bcl-2表达均显著高于对照组,低氧预适应组又显著高于低氧组。低氧组和低氧预适应组Caspase-3表达均显著高于对照组,但低氧预适应组显著低于低氧组。结果表明,低氧预适应过程中,小鼠皮层脑区通过Bcl-2高表达和Caspase-3低表达抵御皮层细胞凋亡,从而参与脑保护机制。关键词:低氧预适应;Bcl-2;Caspase-3;凋亡;皮层;小鼠中图分类号:R364.2C

5、.FluorescenceintensityofBcl-2andCaspase-3wasobservedandnumberofpositivecellswascountedinparietalcortexbyimmunofluorescenceandconfocallaserscanningmicroscope.FluorescenceintensityofBcl-2inthenormalgroup,hypoxicgroupandhypoxicpreconditioninggroupwas6.2±1.7,
6、68.5±13.1,180.6±34.8,respectively,andnumberofBcl-2-positivecellswas18.5±4.9,52.3±10.5,150.8±24.7,respectively.FluorescenceintensityofCaspase-3inthecontrolgroup,hypoxicgroupandhypoxicpreconditioninggroupwas8.6±2.0,40.2±8.2,26.4±6.1,respectively,andnumberof
7、Caspase-3-positivecellsofwas4.3±1.2,63.6±12.5,45.7±9.8,respectively.TheresultsshowedthattheexpressionsofBcl-2inbothhypoxicgroupandhypoxicpreconditioninggroupweresignificantlyhigherthanthatinthecontrolgroup;andtheexpressionofBcl-2inhypoxicpreconditioninggr
