3、age.Thisconclusionisbasedonapronouncedrestrictionfragmentlengthpolymorphism(RFLP)detecteduponhybridizationwithinternalfragmentsofeightresidentinsertionsequences(IS).GeneticdiversitywasdependentontheISconsideredand,inmanycases,aclearconsequenceofIStransposition.IS5wasparticularlyactiveintheg

4、enerationofvariation.AllsubclonesinwhichIS30hadbeenactivetestifytoaburstofIS30transposition.Thiswascorrelatedwithalossofprototrophyandareducedgrowthonrichmedia.ApedigreeoftheentireclonecouldbedrawnfromtheRFLPpatternsofthesubclones.Outof118subclonesanalyzed,68differentpatternswerefoundbutthe

5、putativeancestralpopulationhaddisappeared.Afewpatternswereeachrepresentedbyseveralsubclonesdisplayingimprovedfitness.TheseresultsofferinsightsintotheroleofISelementsintheplasticityoftheE.coligenome,andtheyfurtherdocumentthatenzyme-mediatedDNArearrangementsdooccurinrestingbacterialcultures.T
