1、平颤颗粒治疗帕金森病的药效学研究【摘要】目的:观察平颤颗粒对帕金森病(PD)模型鼠的行为及多巴胺能神经元的影响并探讨其治疗PD的作用机制.方法:运用6羟基多巴胺两点注射法损毁大鼠右侧黑质致密部(SNC)和腹侧被盖区(VTA)多巴胺能神经元,制备PD模型,将模型成功大鼠随机分为模型组、美多芭组、平颤颗粒低、中、高剂量3组以及联合用药组,另设正常对照组,每组10只,分别给予相应处理,连续5inergicneuroninParkinsondisease(PD)modelratsandexplorethepharmacodynamics

3、tenratsalcontrolgroup.Allratsentbyapomorphineberoftyrosinehydroxylase(TH)positivecellsmunohistochemicalmethod,anddopamine(DA)anditsmetabolicproductsinnigrostriatalsystemofeachgroupinedbyusinghighperformanceliquidchromatographycoupledtoelectrochemicaldetection(HPLCEC
4、D).RESULTS:paredodelgroup,thetimesofratsrotationentinthemadopargroupandbinedgroup(P<0.05);thetimesofratsrotationdidntdecreaseinPingchanPotion16g/kgand8g/kggroup,butitdidnotchangeobviouslyin4g/kggroup(P>0.05);thetimesofratsrotationinmodelgroupincreasedposttreat
5、meat(P<0.05),andthebehaviourofnormalgroupdidntchange.ThenumberofTHpositivecellsinsubstantialnigraofratstreatedodelgroup(P<0.05),yetloalgroup.ThelevelsofDAand3,4dihydroxyphenylaceticacid(DOPAC)intheratsofPingchanPotion(16,8g/kg)andmadoparandbinedgroupsodelgroup
6、(P<0.05),butloalgroup,thelevelsofDAandDOPACinPingchanPotion4g/kggroupdidnotsignificantlychange.CONCLUSION:PingchanPotioncouldpartiallyimprovetherotationalbehaviorinPDmodelrats,andprovideaprotectiveeffectagainst6OHDAinducedneuronaldeathinparkinsonianmodelbyreducin
7、gDAneuronlossandincreasingDAcontent. 【Keyonooxygenase;dopamine 0引言 帕金森病(Parkinson′sdisease,PD)是中老年人常见的神经系统变性疾病,其主要病理改变是中脑腹侧、特别是黑质致密部多巴胺(DA)能神经元进行性变性缺失,导致神经递质DA匮乏.由于PD的发病机制尚不清楚,临床治疗仍以多巴制剂替代为主,但其长期应用会出现明显的副作用或不同程度的药效减退,并可加速PD的自然病程[1].平颤颗粒为纯中药制剂,以往临床研究表明其能有效地改善PD患者的运动
8、症状和精神症状[2],且未发现明显的毒副作用.我们采用6羟基多巴胺(6OHDA)制备PD大鼠模型,观察平颤颗粒对PD模型鼠的行为及黑质DA能神经元的影响,探讨其治疗PD的疗效机制,从而为临床应用提供用药依据. 1材料和方法 1.1材料清洁级
