4、rallydesignatedasrl3(t)G1andrl3(t)G2,wereservedasmaterialsforexploringthemechanismunderlyingtherollingleafcharacteristic.Morphologicalanalysisshowedthat,thesetwomutantshavetypicallyadaxiallyrolledleaves.Inaddition,whencomparedwithwildtype93G11,theplantheightsandpaniclelengthsofrl3(t)G1a
5、ndrl3(t)G2significantlydecreased,aswellastheseedsettingpercentage.Cytologicalanalysissuggeststhattherolledleafphenotypemaybecausedbythechangeofnumberandsizeofbulliformcells.Geneticanalysisindicatedthatrolledleafcharacteriscontrolledbyarecessivenucleargene.Crossingbetweenthetwomutants,th
6、eF1[rl3(t)G1/rl3(t)G2]plantsexhibitrolledleaf,suggestingtheyareallelic.TomaptheRL3(t)gene,anF2populationwasgeneratedbycrossingtherl3(t)G1mutantswithWuyunjing8asamappingpopulation.Byusingsimplesequencerepeat(SSR)markersandsomenewdesignedsequencetaggedsite(STS)markers,RL3(t)wasinitiallyma
7、ppedintheregionbetweentheSTSmarkerM3G45andtheSSRmarkerRM6676withthegeneticdistancesof5.5cMand4.4cM,respectively,onthelongarmofchromosome3.Furthermore,withtheenlargedpopulationandmoredevelopedSTSmarkers,RL3(t)genewasfinallydelimitedtoa46GkblongregiongovernedbytheSTSmarkersS3G39a
