2、杨幼苗为材料,采用石蜡切片法,制作微透析探针插入杨树嫩茎后,对嫩茎受损部分横切面组织切片进行显微观察并加以探讨。结果表明,微透析探针的粗度约为杨树细胞直径20倍,探针对周围细胞造成一定程度的破坏,因此,探针插入杨树嫩茎后开始一段时间内收集的透析液将受到破损细胞液的污染,随着微透析时间的推移,逐渐地趋近于质外体汁液。对微透析技术可以获得植物质外体汁液从细胞显微结构变化方面予以解释,为微透析技术在植物质外体研究中的应用奠定细胞学基础。关键词:微透析;杨树;组织切片;细胞破损;显微观察中圈分类号:$718.4;$792.11文献标识码:A文章编号:1002—2481(2012)
