1、毕业论文(设计)全蝎入药工艺的优化与蝎体蛋白的分离不要删除分节符,此行不会被打印。摘要沂蒙全蝎是我国的名贵药材,其入药对多种疾病有预防和治疗作用。本实验对传统的全蝎炮制、煎煮入药工艺进行改进,并对活蝎蝎体蛋白进行了分离纯化。全蝎的炮制作为传统的制药技术在很大程度上决定着全蝎药材的质量。本实验采用薄层色谱法对全蝎炮制技术进行了优化,得出将全蝎放入清水或2%淡盐水中浸泡4h,然后放入约10-20倍量沸开水中煮沸5min的最优工艺。全蝎煎煮工艺的优化是以蝎毒蛋白的含量为指标,采用考马斯亮蓝G-250染色法测定其含量,用正交设计法进行提取工艺条件的筛选,最终得出了将
2、全蝎粉末用14倍量水浸润15分钟,隔水煮沸60min的最佳工艺条件。采用水提法获得活蝎蝎体的总蛋白,通过SephadexG-75凝胶色谱和SDS-PAGE,得到活蝎蝎体各蛋白组分。与传统的入药蝎体及蝎尾毒蛋白做比较,可知活蝎蝎体比入药蝎体多出分子量为46000和8000的两个蛋白组分,并具有蝎尾毒所缺乏的大分子蛋白。这一结果可以为蛋白质药物研究提供一组基础数据,并对全蝎综合利用方案的提出具有理论指导作用。关键词:沂蒙全蝎,正交设计,蝎体蛋白,分离纯化,凝胶层析AbstractYimengScorpionisakindofraremedicinalherbs.
