1、2013—年第16卷第12期PracticalPharmacyAndClinicalRemedies,2013Vo1.16.No.12·论著·丙戊酸联合顺铂对人肝癌细胞的增殖抑制作用研究蒋凤琴,袁玉梅,汪荣华,姚冲,曹恒斌[摘要]目的探讨丙戊酸(VPA)联合顺铂(DDP)对人肝癌细胞HepG2、BEC7404、SMMC7721的增殖抑制作用。方法三组浓度的VPA与DDP单药及联合作用于人肝癌细胞HepG2、BEC7404、SMMC7721.24、48、72h后,观察细胞数量和形态的变化。用MTT法分析药物对
2、细胞增殖的抑制率(IR),计算联合用药对细胞增殖抑制率q值,考察联合使用是否为协同作用。结果72h后,各用药组细胞数目减少十分明显,其中vPA+DDP组较单独用药组减少更为显著,其细胞形态发生较大改变。采用MTT比色法显示,对照组细胞生长率明显高于各实验组,联合用药组对细胞增殖IR更为显著,且随着药物浓度增加及作用时间延长,各组对细胞增殖IR明显增加。根据q值结果,在VPA较高浓度(≥100g/mE)时,联合用药组具有明显的协同作用。结论VPA能增强DDP对人肝癌细胞的增殖抑制作用,并且在VPA较高浓度时,
