1、临床放射学杂志2014年第33卷第2期·293·实验研究USPIO联合MR活体动态示踪技术在EPCs移植脑卒中大鼠模型中的作用探讨侯唯姝,徐丽艳,余永强【摘要】目的证实超微超顺磁性氧化铁颗粒(USPIO)作为细胞内对比剂标记大鼠骨髓源性内皮祖细胞(EPCs)的可行性,并以大鼠大脑中动脉阻塞(tMCAO)模型为载体,探讨MR动态示踪在干细胞移植在中的作用。方法多聚赖氨酸(PLL)为转染剂、USPIO为细胞内对比剂标记EPCs,于第1、3、5天分别计算细胞标记率并检测细胞活力。tMCAO大鼠3O只,随机分为两组,分别经尾静脉移植磁性标记EPCs(
2、实验组)及未标记EPCs(对照组),行MR动态扫描及普鲁士蓝染色。结果USPIO—PLL能够成功标记EPCs,标记率达98%,标记组与未标记组间细胞活力无明显差异。MRTWI序列动态扫描于脑梗死区与正常脑组织交界处的边缘可观察到连续的低信号,Tmap序列更为明显。普鲁士蓝染色结果与体外细胞染色结果相符。结论USPIO可以成功标记EPCs,MR活体动态示踪能够成功监测到磁性标记细胞。【关键词】超小超顺磁性氧化铁磁共振成像内皮祖细胞脑卒中ApplicationofUltrasmallSuperparamagneticIronOxidePartic
