1、144ChineseJournalofClinicalMedicine,2014.Vo1.21,No.2中国临床医学2014年4月第21卷第2期论著大豆异黄酮对大鼠心肌缺血再灌注损伤的保护作用何悦王浩曲毅(1,上海市徐汇区中心医院老年科,上海200031;2.复旦大学上海医学院基础医学实验中心,上海200032)摘要目的:探讨大豆异黄酮(soybeanisoflavone,SI)对大鼠心肌缺血再灌注损伤的保护作用及其可能的机制。方法:采用结扎冠状动脉法构建大鼠心肌缺血再灌注模型。按照随机数字表法将100只雄性SD大鼠随机分为4组:假手术组(S
2、ham组)、缺血再灌注组(I/R组)、SI处理组(SI组)、SI+JAK激酶抑制剂AG490组(SI+AG490组),每组25只。采用原位末端标记法(TdT—mediateddUTPnickend1abeling,TUNEL)检测心肌细胞的凋亡;采用免疫组织化学法检测凋亡相关因子Bax及Bcl-2蛋白的表达;采用蛋白质印迹法(Westernblotting)检测Janus蛋白酪氨酸激酶2(Janusactivatedkinase2,JAK2)、磷酸化JAK2及信号转导与转录激活因子3(signaltransducerandactivatoro
