1、广西农业科学2010,41(8):758—761GuangxiA鲥culturMSciences山薯组培苗继代培养基配方筛选严华兵h,龚明霞·b,董伟清h,1-朝阳la)闭志强h,李杨瑞1(1广西农业科学院a生物技术研究所;b蔬菜研究所,南宁530007)摘要:以山薯组培苗带腋芽茎段为外植体,以Ms为基本培养基,探讨植物生长调节剂6—8A(0.5、1.0、2.0rag/L)或TDZ(0.01、0.10、0.50、1.00、2.00、4.00mg/L)、不同培养方式和活性炭对其继代培养的影响。结果表明,添加不同浓度6一BA对山薯组培苗增殖系数、茎长及腋芽点芽球
2、的影响不明显,而随着TDZ浓度的升高,山薯组培苗增殖系数和茎长逐渐降低而芽球显著增加,茎段腋芽生长受到明显抑制。山薯组培苗较优的继代培养基为MS+I.0mg/L6-BA+0.1mg/LNAA+30g/L蔗糖+1.5g/L活性炭的液体培养基。液体培养基利于促进山薯组培苗增殖。其增殖系数、茎长、植株鲜重和干重显著高于固体培养基。活性炭的添加可以减少褐化,促进组培苗的增殖培养。此外,培养方式与活性炭交互作用对组培苗茎长、增殖系数、植株鲜重及干重无显著影响。关键词:山薯;继代培养;增殖系数;固体培养;液体培养;活性炭中图分类号:$632.103".53文献标识码:A
