1、·986·ChinJDisControlP口2014Oct;18(10大豆异黄酮对高胆固醇模型大鼠血清胆固醇浓度和肝脏胆固醇代谢的影响徐超,张卓,王晓红,周波,郭连营【摘要】目的观察大豆异黄酮对高脂模型大鼠血清胆固醇浓度及其肝脏胆固醇代谢关键酶基因表达的影响。方法9周龄SD雄性大鼠8只喂饲AIN93M饲料(阴性对照组),18只喂饲高脂饲料建立高胆固醇模型。14d后将高胆固醇大鼠随机分为异黄酮组(每天灌胃360mg/kgBW大豆异黄酮)和高脂对照组,喂饲高脂饲料至实验结束。30d后测血脂,实时定量聚合酶链式反应(polymerasechainre
2、action,PCR)测定肝脏羟甲戊二酰辅酶A还原酶(3-hydroxy一3一methylglutaryl—CoenzymeAreductase,Hmgcr)、胆固醇7·羟化酶(eytochromeP450,family7,subfami—lya,polypeptide1,Cyp7a1)、低密度脂蛋白受体(1owdensitylipoproteinreceptor,Ldlr)mRNA表达量。结果异黄酮组大鼠血清总胆固醇(totalcholesterol,TC)和低密度脂蛋白胆固醇(1owdensitylipoproteincholesterol
3、,LDLC)浓度分别比高脂对照组降低了23.7%(q=2.79,P=0.010)和23.2%(q=2.63,P=0.015);动物肝脏的Hmgcr和Cyp7a!mRNA表达量均出现下降,但差异无统计学意义(均有P>0.05)。结论大豆异黄酮可明显降低高胆固醇大鼠血清TC和LDLC浓度,有可能影响肝脏胆固醇代谢。【关键词】大鼠;胆固醇;异黄酮类【中图分类号】R344;R977.6【文献标识码】A【文章编号】1674-3679(2O14)10-0986-05Efectofsoyisoflavoneontheserumcholesterolconce
