1、Chapter11AnRT-PCRAssayforDetectionofInfectiousBronchitisCoronavirusSerotypesJunfengSunandShengwangLiuAbstractAvianinfectiousbronchitisvirus(IBV),achickenGammacoronavirus,isamajorpoultrypathogen,andisprobablyendemicinallregionswithintensivepoultryproduction.SinceIBVwasÞrstdescribedin1936,manyserotype
2、sandvariantsofIBVhavebeenisolatedworldwide.IBVisolatesarecapableofinfectingalargerangeofepithelialsurfacesofthechicken,involvingtherespiratory,renal,andreproductivesystems;however,theclinicalsignsareusuallynotspeciÞcfordifferentialdiagnoses.VirusisolationiscommonlyusedfordiagnosisofIBVinfection,whic
3、hwasachievedthroughpassageofclinicalmaterialsviatheallantoicrouteofembryos.Currently,moresensitivemolecularapproachesforthedetectionofavianpathogenshavebeendeveloped,includingreverse-transcriptasepolymerasechainreaction(RT-PCR)andreal-timeRT-PCR,whicharemoresuitableforuseindiagnosticlaboratories.Int
4、hischapter,wedescribeaone-stepRT-PCRwhichcanbeusedfordetectingmostofIBVserotypesintheIBV-infectedallantoicßuidandhasbeenusedroutinelyinourlaboratoriesfordetectionofIBVs.KeywordsIBV,RT-PCR,Moleculardetection,Diagnosis,Clinicalsamples1IntroductionInfectiousbronchitis(IB)isanacuteandhighlycontagiousvir
5、aldiseaseofchickencausedbytheinfectiousbronchitisvirus(IBV).Thisdiseaseisprevalentthroughouttheworldandaffectstheperformanceofbothmeat-typeandegg-layingbirds,therebycausingsevereeconomiclosswithinthepoultryindustry.IBcanaffectchickensofallagesandresultinrespiratorydisease,nephritis,proventriculitis,
6、anddecreaseineggproductionandeggquality[1Ð3].Furthermore,thisdiseaseisoftencomplicatedbysecondarybacterialinfectionsthatcauseincreasedmortality.Itmaybethemosteconomicallyimportantviralrespiratorytractdiseaseofchickensincountriesandregionswherethereisnohighlypathogenicavianinßuenzavirusorvelogenic(hi
