3、wasnotusedinthemaintenancegation.Nineofthe23coronavirusstrainsmedium.recoveredtodatewereoriginallyisolatedinInfectivitytitrations.Tenfolddilutionsofhumanembryonictracheal(HET)organcul-virusweremadeinHanks'BSScontainingturesonly,whereastheremaining14,allof0.5%gelatin,100unitsofpenicillinJm1andwhichw
4、eresimilartoaprototypestrain100,pgstreptomycin/ml.Two-tenthsmilliliterdesignated2293,wereisolatedwithdifficultyoftheappropriatevirusdilutionswasinocu-inmonolayertissuecultures(2,4-9).latedintoeachtubeculture;theculturesB814virus,thefirstorganculture(OC)wereexaminedforhemadsorptionasde-strain,twooth
5、erOCstrains,LPandEVS,scribedbelowatvaryingintervalsundertheandstrain2293weresubsequentlyrecoveredlowpowerofalightmicroscope.IfculturesinL132cellmonolayersfromoriginalnasalweretobeexaminedagainforhemadsorptionwashingsobtainedfromvolunteersinoculatedatalaterdate,themaintenancemediumcon-withtheseagent
6、s(8,10).Morerecently,tainingredbloodcellswasdecantedandtheOC43virus(andOC38virus,anidenticalmonolayerwaswashedtwicewithHanks'BSSagent)bothofwhichwereoriginallyrecov-beforetheadditionofnewmaintenanceeredinOCandlateradaptedtogrowinmedium.sucklingmice,werenotonlyshowntodirect-Serology.Allserawereinact
