3、antpathogensresponsibleforrespira-tory,gastrointestinalandpossiblyneurologicaldisorders.TobetterunderstandthemolecularbiologyoftheprototypeHCV-229Estrain,thenucleotidesequenceofthe5-uniqueregionsofmRNAs4and5weredeterminedfromclonedcDNAs.SequenceanalysisofthecDNAssynthesizedfrommRNA4re
4、vealedamajordifferencewithpreviouslypublishedresults.However,polymerasechainreactionamplificationofthisregionshowedthatthesequencedcDNAswereproducedfromminorRNAspecies,anindicationofpossiblegeneticpolymorphisminthisregionoftheviralgenome.ThemutatedmessengerRNA4containstwoORFs:(1)ORF4a
5、consistingof132nucleotideswhichpotentiallyencodesa44-aminoacidpolypeptideof4653Da;thiscodingsequenceisprecededbyaconsensustranscrip-tionalinitiationsequence,CUAAACU,similartotheonesfoundupstreamoftheNandMgenes;(2)ORF4bof249nucleotidespotentiallyencodingan83-aminoacidbasicandleucine-ri
6、chpolypeptideof9550Da.Ontheotherhand,mRNA5containsonesingleORFof231nucleotideswhichcouldencodea77-aminoacidbasicandleucine-richpolypeptideof9046Da.Thisputativeproteinpresentsasignificantdegreeofaminoacidhomology(33%)withitscounterpartfoundintransmissiblegastroenteritiscoronavirus(TGEV
