4、efecesandnasalsecretionsofinfectedcalves.Fecesornasalswabfluidswereincubatedsequentiallywithhyperimmunebovineanti-bovinecoronavirusserumandproteinA-gold,negativelystained,appliedtoformvar-coatedcoppergridsandviewedusinganelectronmicroscope.ThePAG-IEMmethodspecificallyid
5、entifiedBCVparticlesandpossiblesubviralparticlesinfecesandnasal-swabfluidsfrominfectedcalves.ThePAG-IEMmethoddidnotlabelotherenvelopedentericvirusesormorphologicallysimilarfringedparticlescommonlyfoundinfeces.DetectionofBCVusingPAG-IEMwascomparedwithELISAanddirectimmuno
6、fluorescence(IF)ofnasalepithelialcellsbymonitoringfecalandrespiratorytractsheddingofBCVfromtwoexperimentallyinfectedandtwonaturallyinfectedcalvesfrombirthto3weeksofage.PAG-IEMandELISAdetectedsheddingofBCVinfecal(4/4animals)andnasal(3/4animals)samplesforanaverageof5.25da
7、yseach.TheobservedagreementofBCVdetectionbyPAG-IEMandELISAwas85%.PAG-IEMmaybeamoresensitiveimmu-noassayforthedetectionofBCVindiagnosticspecimensfrominfectedneonatalcalvesthanELISA.BCVinfectionofnasalepithelialcellswasdetectedbyimmunofluorescencein4/4calves,persistedfort
