2、能,筛选合适的ProteinA填料,用于大规模单克隆抗体(简称单抗)纯化。方法采用表达H02单抗的CHO细胞培养上清,测试不同供应商ProteinA填料的蛋白结合载量,选择合适的填料,用于大规模单抗纯化;检测宿主细胞蛋白(hostcellprotein,HCP)、外源DNA及ProteinA残留量,以评价其纯化效果;检测ProteinA柱在位清洗(cleaninginplace,CIP)效果,并验证其循环使用次数。结果AmsphereProteinAJWT203填料为合适的ProteinA填料,H02单抗大规模纯化后,可
3、有效去除HCP、DNA及脱落ProteinA;采用0.1mol/LNaOH进行CIP,经过300个循环,对H02单抗仍保持80%以上动态结合载量,ProteinA脱落无明显增加,对HCP、外源DNA、聚合体的去除能力无明显改变,层析图谱与初始基本一致,AmsphereProteinAJWT203填料的理化性质较稳定。结论AmsphereProteinAJWT203是一种可用于大规模单抗纯化的新型ProteinA填料。关键词:抗体,单克隆;纯化;ProteinA填料中图分类号:O657.7+3R392-33文献标识码:A文
