3、eonthereliableassessmentoflipidperoxidation.Malondialdehyde(MDA)isoneofseverallow-molecular-weightendproductsformedviathedecompositionofcertainprimaryandsecondarylipidperoxidationproducts.AtlowpHandelevatedtemperature,MDAreadilyparticipatesinnucleophilicadditionre
4、actionwith2-thiobarbituricacid(TBA),generatingared,fluorescent1:2MDA:TBAadduct.Thesefacts,alongwiththeavailabilityoffacileandsensitivemethodstoquantifyMDA(asthefreealdehydeoritsTBAderiva-tive),haveledtotheroutineuseofMDAdeterminationand,particularly,the"TBAtest"to
5、detectandquantifylipidperoxidationinawidearrayofsampletypes.However,MDAitselfparticipatesinreactionswithmoleculesotherthanTBAandisacatabolicsubstrate.OnlycertainlipidperoxidationproductsgenerateMDA(invariablywithlowyields),andMDAisneitherthesoleendproductoffattype
6、roxideformationanddecompositionnorasubstancegeneratedexclusivelythroughlipidperoxidation.Manyfac-tors(e.g.,stimulusforandconditionsofperoxidation)modulateMDAformationfromlipid.Additionalfactors(e.g.,TBA-testre-agentsandconstituents)haveprofoundeffectsontestrespons
7、etofattyperoxide-derivedMDA.TheTBAtestisintrinsicallynonspecificforMDA:nonlipid-relatedmaterialsaswellasfattyperoxide-deriveddecompositionproductsotherthanMDAareTBApositive.TheseandotherconsiderationsfromtheextensiveliteratureonMDA,TBAreactivity,andoxidativelipidd
8、egradationsupporttheconclusionthatMDAdeterminationandtheTBAtestcanoffer,atbest,anarrowandsomewhatempiricalwindowonthecomplexprocessoflipidperoxidation.T
