3、mentalendstateofplantcells.TwotypesofMYBtranscriptionfactors,R2R3-MYBandR3-MYB,areinvolvedincellfatedetermination.Toexaminethemolecularbasisofthisprocess,weanalyzedthefunctionalrelationshipoftheR2R3-typeMYBgeneWEREWOLF(WER)andtheR3-typeMYBgeneCAPRICE(

4、CPC).ChimericconstructsmadefromtheR3MYBregionsofWERandCPCusedinreciprocalcomplementationexperimentsshowedthattheCPCR3regioncannotfunctionallysubstitutefortheWERR3regioninthedifferentiationofhairlesscells.However,WERR3cansubstantiallysubstituteforCPCR3

5、.TherearenodifferencesinyeastinteractionassaysofWERorWERchimeraproteinswithGLABRA3(GL3)orENHANCEROFGLABRA3(EGL3).CPCandCPCchimeraproteinsalsohavesimilaractivityinpreventingGL3WERandEGL3WERinteractions.Furthermore,weshowedbygelmobilityshiftassaysthatWE

6、RchimeraproteinsdonotbindtotheGL2promoterregion.However,aCPCchimeraprotein,whichharborstheWERR3motif,stillbindstotheGL2promoterregion.INTRODUCTIONtouchingonlyonecorticalcelldevelopintohairlesscells.Wild-typeArabidopsishaseighthaircellfilesalignedlongit

7、udinallyCellfatedeterminationisacriticalstepinthedevelopmentalalongtheroot.processesofplantsandinvolvestheparticipationofalargeFrompreviousworkitisclearthattwotypesofMYB-relatednumberoftranscriptionfactors.TheMYBfamilyisoneofthetranscriptionfactors(R2

8、R3andR3)areinvolvedinepidermisdif-largestgroupsoftranscriptionfactorsintheArabidopsisthalianaferentiation.ThecloselyrelatedWEREWOLF(WER),GLABRA1genome,with>125members(Kranzetal.,1998;Strackeetal.,(GL1),andMYB23genesencodeR2R3-typeMYBgenes.WER2
