3、March16,2004Abstract—Themechanismoftheincreasedaccumulation(overproduction)ofcitricacidsintheyeastYarrowialipolyticawhilegrowinginthepresenceofglucoseundernitrogendeficiencywasinvestigated.Thelimitationoftheyeastgrowthbythesource++ofnitrogendecreasesthetotalcontentofnucleotidesandincreas
4、estheratiosofATP/AMPandNADH/NAD.NADDependentisocitratedehydrogenase,anenzymeofthetricarboxylicacidcycleplayingakeyroleintheregulationofbiosynthesisofcitricandisocitricacids,wasisolatedfromY.lipolytica.Themolecularweightsofthenativeenzymeanditssubunitswerefoundtobe412and52kD,respectivel
5、y.Itisconcludedthattheenzymeisahomooligomerconsistingofeightsubunits.Investigationoftheeffectofsomeintermediatesofthetricarboxylicacidcycleontheactivityofthisenzymesuggeststhatthe+enhancedexcretionofcitricacidscanbecausedbytheinhibitionofNADdependentisocitratedehydrogenaseduetothe+decre
