2、BSTRACT:Wehavedevelopedaconvenientandefficientcolorimetricdetectionsystemforproteintargetsusingaptamer−goldnanoparticleconjugates.Wetakeadvantageofthecorrelationbetweenthecatalyticpropertiesandtheexposedsurfaceareaofthenanoparticles,whichisinverselyproport
3、ionaltotheamountoftheaptamer-boundproteintargets.Astheconcentrationoftheproteintargetincreases,thenanoparticlesurfaceareabecomesmoremasked,thusincreasingthereductiontimeof4-nitrophenolforthecolorchange.Wealsoreducethedetectiontimebyeitherredesigningtheap
4、tamersequencesorregulatingtheirdensity.Thisdetectionsystemishighlyselective,discriminatingthetargetproteinevenataconcentration1000timeshigherthanthelimitofdetection(LOD).Importantly,tothebestofourknowledge,theLODwiththeunaidedeyeinthisworkisthelowestfora
5、colorimetricdetectionsystemusinglysozymeasamodelprotein(16nM).Lysozymeinchickeneggwhitesisdirectlyanalyzedusingourdetectionsystem,whoseresultsareinexcellentagreementwiththeenzyme-linkedimmunosorbentassay(ELISA)analysis.oldnanoparticles(AuNPs)haveservedas
6、the“gold”Ifthesenanoparticles,inadditiontotheircatalyticproper-Gstandardinthefundamentalsandapplicationsofties,31,32couldaccuratelyrecognizeaspecifictargetwithnanoscienceowingtotheiruniquechemicalandphysicalassistancefromsurface-conjugatedchemicalligandss
7、uchas1−5properties.Onthebasisofsuchproperties,anumberofaptamers,33−36theywouldbespecificallydevisedas“artificial”diagnosticapplicationsofAuNPswithhighsensitivityandenzymesforbiosensing.selectivityhavebeendemonstrated,particularlyfortheHerein,wepresentahigh
8、lysensitiveandselectivedetectionofphysiologicallyandclinicallyimportantproteintargets.6−10DependingontheroleofAuNPsinthedetectioncolorimetricdetectionofproteintargetsusingaptamer−gold37scheme,varioustypesofreadoutsareemplo
