3、lts:Byusinganovelhigh-throughputmethod,namedchemicalinferenceofRNAstructures,CIRS-seq,thatusesdimethylsulfate,andN-cyclohexyl-N'-(2-morpholinoethyl)carbodiimidemetho-p-toluenesulfonatetomodifyRNAresiduesinsingle-strandedconformationwithinnativedeproteinizedRNAsecondarystr

4、uctures,weinvestigatethestructuralfeaturesofmouseembryonicstemcelltranscripts.Ouranalysisrevealsanunexpectedhigherstructuringofthe5′and3′untranslatedregionscomparedtothecodingregions,areducedstructuringattheKozaksequenceandstopcodon,andathree-nucleotideperiodicityacrossth

5、ecodingregionofmessengerRNAs.WealsoobservethatncRNAsexhibitahigherdegreeofstructuringwithrespecttoproteincodingtranscripts.Moreover,wefindthattheLin28abindingproteinbindsselectivelytoRNAmotifswithastrongpreferencetowardasinglestrandedconformation.Conclusions:Thisworkdefin

6、esforthefirsttimethecompleteRNAstructuromeofmouseembryonicstemcells,revealinganextremelydistinctRNAstructurallandscape.TheseresultsdemonstratethatCIRS-seqconstitutesanimportanttoolfortheidentificationofnativedeproteinizedRNAstructures.Backgroundnuclearribonucleicparticle(

7、snRNP)complexes[9,10],Thedevelopmentofhigh-throughputmethodsforthemostncRNAsarethoughttoexerttheirfunctionsbyanalysisoftheepigenomeandtranscriptomehaveledtofoldingintolocallystablesecondarystructuresthatmaythediscoveryofthousandsofpreviouslyunannotatedprovideanchoringsite
