2、amyloidfibrilsformedbyisletamyloidpolypeptide(IAPP)areassociatedwithtypeIIdiabetes.OneoftheproposedmechanismsofthetoxicityofIAPPisthatitcausesmembranedamage.ThefatalmutationofS20GhumanIAPPwasreportedtoleadtoearlyonsetoftypeIIdiabetesandhightendencyof

3、amyloidformationinvitro.CharacterizingthestructuralfeaturesoftheS20Gmutantinitsmonomericstateisexperimentallydifficultbecauseofitsunusuallyfastaggregationrate.Computationalworkcomplementsexperimentalstudies.Weperformedaseriesofmoleculardynamicssimula

4、tionsofthemonomericstateofhumanvariantsinthemembrane.Oursimulationsarevalidatedbyextensivecomparisonswithexperimentaldata.WefindthatahelicaldisruptionatHis18iscommontobothhumanvariants.AnL-shapedmotifofS20Gmutantisobservedinoneoftheconformationalfami
