3、-SNPanalysisoftenidentifiesonlyafewofthemostsignificantSNPsthataccountforasmallproportionofthegeneticvariantsandoffersonlyalimitedunderstandingofcomplexdiseases.Toovercometheselimitations,weproposegeneandpathway-basedassociationanalysisasanewparadigmforGWAS.Asaproofofconcep
4、t,weperformedacomprehensivegeneandpathway-basedassociationanalysisof13publishedGWAS.OurresultsshowedthattheproposednewparadigmforGWASnotonlyidentifiedthegenesthatincludesignificantSNPsfoundbysingle-SNPanalysis,butalsodetectednewgenesinwhicheachsingleSNPconferredasmalldiseas
5、erisk;however,theirjointactionswereimplicatedinthedevelopmentofdiseases.TheresultsalsoshowedthatthenewparadigmforGWASwasabletoidentifybiologicallymeaningfulpathwaysassociatedwiththediseases,whichwereconfirmedbyagene-set-richanalysisusinggeneexpressiondata.EuropeanJournalof
