4、erformed.Results暋TheexpressionofEphA1transcript,methylatedDNAofE灢phA1wasdetectedincelllines.EphA1mRNAexpressioninthecorrespondingnormalmucosaeisnotsignificantlydifferentfromthatintumortissues(P=0.496).Inadenomas,theimmunostainingforEphA1waspositivediffuselyandstrongly.However,inCRC
5、s,theexpressionofEphA1proteinwaspredominatelydown灢regulated.ReducedorlostexpressionoftheEphA1proteinwassignificantlyas灢sociatedwithpoordifferentiation(P=0.027),greaterdepthofwallinvasion(P=0.002)andad灢vancedTNMstage(P=0.002),andpreferentiallyoccurredinpatientswithlymphnodemetastasi
6、s(P=0.019).PatientswithreducedEphA1expressionhadatendencyofpoorerprognosisthanthosewithEphA1upregulation.Concolusion暋EphA1mayhavetumorsuppressorrolesinCRCsandmaybeanimportantbiomarkerbywhichmalignancy,biologicalbehaviorandprognosisareevaluated.Keywords:EphA1;Receptortyrosinekinase;
7、Colorectalneoplasm;Real灢timeRT灢PCR;Methylation摘暋要:目的暋探讨EphA1在结直肠癌中的表达及其临床意义。方法暋利用荧光实时定量RT灢PCR检测EphA1在肠癌细胞系和组织中的表达,并分析甲基化状态。免疫组织化学染色分析E灢phA1蛋白在肠癌组织中的表达及其临床意义。结果暋EphA1在肠癌细胞中不同程度表达,并存在甲基化。EphA1mRNA在癌旁正常黏膜和肿瘤组织中表达差异无统计学意义(P=0.496)。EphA1蛋白在肠腺瘤中弥漫强阳性,在癌组织中主要为低或缺失表达,低表达多见于低分化腺癌(P=0.027)、伴
