3、fectsofplateletactivatingfactor(PAF)ontheactionpotentialandpotassiumcurrentsinguinea-pigventricularmyocytes.Wholecellpatchclamptechniqueswereused.With5mmol/LATPinthepipetteelectrode(mimicnormalcondition),1µmol/LPAFincreasedAPDfrom225.8±23.3to352.8±29.8ms(n=5,P<0.05)
4、,decreasedIandItail90K1Kcurrentsfrom-6.1±1.3to-5.6±1.1nA(n=5,P<0.05)at-120mVandfrom173.5±16.7to152.1±11.5pA(P<0.05,n=4)at+30mV,respectively.ButPAFhadnoeffectonIatpotentialswithinthenormalrangeofmembranepotentials(between-90mVand+20mV).K1Inthecontrary,withoutATPinthe
5、pipetteelectrodebywhichIwasactivated(mimicischemiccondition),1µmol/LPAFshortenedK·ATPAPDfrom153±24.6to88.2±19.4ms(n=5,P<0.01).Incubationofmyocyteswith1µmol/Lglibenclamide,ablockerofIcould90K·ATPrestoreprolongationofAPDinducedbyPAF.Inconclusion,inguinea-pigventricula
6、rmyocytes,with5mmol/LATPinthepipettePAFcouldprolongAPDpartlyduetotheinhibitionofI;whilewith0mmol/LATPinthepipette,PAFcouldinduceanactivationofI,KK·ATPhenceadecreaseinAPD.ItissuggestedthatPAFmayamplifytheheterogeneitybetweenischemicandnormalcardiacmyocytesduringische
