2、Lanzhou,ChinaAbstractBackground:Viciasativa(thecommonvetch)possessesapredominantzygomorphicflowerandbelongstothesubfamilyPapilionoideae,whichisrelatedtoArabidopsisthalianaintheeurosidIIcladeofthecoreeudicots.Eachvetchflowerconsistsof21concentricallyarrangedorgans:theoutermostfivesepals,t
3、henfivepetalsandtenstamens,andasinglecarpelinthecenter.Methodology/PrincipalFindings:Weexploredthefloraltranscriptometoexamineagenome-scalegeneticmodelofthezygomorphicflowerofvetch.mRNAwasobtainedfromanequalmixtureofsixfloralorgans,leavesandroots.Denovoassemblyofthevetchtranscriptomeusin
4、gIlluminapaired-endtechnologyproduced71,553unigeneswithanaveragelengthof511bp.Wethencomparedtheexpressionchangesinthe71,553unigenesintheeightindependentorgansthroughRNA-SeqQuantificationanalysis.Wepredominantlyanalyzedgeneexpressionpatternsspecifictoeachfloralorganandcombinationsoffloral
5、organsthatcorrespondedtothetraditionalABCmodeldomains.ComparativeanalyseswereperformedinthefloraltranscriptomesofvetchandArabidopsis,andgenomesofvetchandMedicagotruncatula.Conclusions/Significance:OurcomparativeanalysisofvetchandArabidopsisshowedthatthevetchflowersconformtoastrictABCmode
6、l.WeanalyzedtheevolutionandexpressionoftheTCPgenefamilyinvetchatawhole-genomelevel,andseveralunigenesspecifictothreedifferentvetchpetals,whichmightoffersomecluestowardelucidatingthemolecularmechanismsunderlyingfloralzygomorphy.Ourresultsprovidethefirstinsightsintothegenome-scalemolecular
