1、硕士学位论文中文摘要中文摘要目的观察大鼠坐骨神经切断立即缝合术后不同时间点降钙素莲固扭差肽(CalcitoninGene—relatedPeptide,CGRP)在背根节与脊髓中表达的变化及应用外源性神经生长因子(NerveGrowthFactor,NGF)后对其表达的影响。方法成年SD雄性大鼠110只,随机分为实验组、实验对照组、假手术对照组(各35只)与正常对照组(5只)。实验组动物右侧坐骨神经切断后立即缝合,经腹腔注射每天给予NGF(400单位/kg),实验对照组动物在右侧坐骨神经切断立即缝合后经腹腔注射每天给予等量生理盐水(Ns)
2、;假手术组动物仅暴露坐骨神经,不做神经切断与缝合;动物分别存活1,3,5,7,14,21,28天,免疫组织化学方法结合图像分析技术检测与坐骨神经相连的背根节(DorsalRootGanglion,DRG)和脊髓中CGRP的表达;正常对照组动物不作任何处理。结果正常大鼠DRG与脊髓前角内有明显的CGRP免疫阳性神经元,脊髓后角浅层有明显的CGRP免疫阳性神经纤维。假手术组的结果与正常组基本一致,两组CGRP免疫阳性神经元或神经纤维平均光密度值比较差异没有显著性(P>0.05)。实验对照组大鼠:术后1天右侧DRG内CGRP的表达与正常组和假手
