1、※基础研究食品科学2011,Vo1.32,No.1933苦荞多糖的分离纯化及单糖组成测定颜军1,孙晓春2,谢贞建1,何钢1,易勇1,房晨1,苟小军II21(1.成都大学中药化学实验室,四川成都610106;2.四川抗菌素工业研究所,四川成都610052)摘要:采用水提醇沉法获得苦荞多糖,经离子交换柱分离,高效凝胶过滤色谱法(HPGFC)纯度鉴定并测定多糖的相对分子质量。硫酸水解后,用1一苯基.3.甲基一5.吡唑啉酮(PMP)衍生水解后的单糖,衍生物采用反相高效液相色谱法测定单糖组成。结果表明:苦荞经水提醇沉,用Sevag法脱蛋白后,获得苦荞多糖(TBP);DEAE.纤维素
2、柱层析获得3个苦荞多糖组分TBP一1、TBP2和TBP一3,相对分子质量分别为144544、445656和636795。柱前衍生液相色谱分析可知:TBP.1、TBP.2是由葡萄糖组成的均一多糖;TBP.3由甘露糖、鼠李糖、葡萄糖醛酸、葡萄糖、半乳糖、阿拉伯糖组成的杂多糖,其物质的量比为4.32:2.41:1.00:39.8:9.64:2.02。关键词:苦荞;多糖;单糖;衍生化;高效液相色谱法PurificationandMonosaccharideCompositionAnalysisofTartaryBuckwheatPolysaccharidesYANJun,SUNX
