3、stigations,suchasisolation,characterizationandstudyofalargeextrachromosomalDNAwerealsotakenplace.Sothedissertationispresentedindifferentstepsbellow.Firstly,ahighlysalttolerantstrainobtainedfromcoastline,WasidentifiedasGracilibacillussp.GTYonthebasisofmorph

4、ologicalandphysio—biochemicalcharacteristicsand16SrDNAsequenceanalysis.AccordingtoDNAsequence,themaximum93%-95%homologywasfoundforthestrainGTY.ThestrainwasdepositedasapatentstraininChinaGeneralMicroorganismCultureCenterwiththeaccessionnumberCGMCC1527.The16

5、SrDNAsequenceofGTYwasalsosubmittedtoGenBankwiththenumberDQ286727.Thestrainwasgram—negative,rod-shapedandstrictlyaerobic.Optimumgrowthconditionswereobserved:temperature+300C,pH7.2,10%一15%(w/v)ofNaCl,andprolineasthebestcompatiblesoluteaswellasagoodcarbonsour

6、ce.Therapidandsmoothgrowthrateswereobservedin10%一15%(w/v)ofNaCIsupplementedLuria·Bertani(LB)andMineral-Salt(MS)media,butthestraincouldsurvivenarrowlyinthemediacontainingupto25%(w/v)ofNaCl.Soitwasconsideredasahighlysalttolerantstrain.Md.SalahUddin:Character
