2、r:micro-patternedSiO2/Siwaferandmicro-patternedAu/SiO2wafer,usedasthesupportsofbiochips.Theirsurfacetreatment,biodetectionfeasibilityandtheirapplicationsinopticaldetectionwereexplored.Theresearchachievementasfollowings:1.Thesurfacetreatmentandmodificationofthemicro-patternedSiO2/Sisiliconwafer.Con
3、centratedH2SO4/H2O2solution,concentratedHCL/H2O2/H2Osolution,NH4OH/H2O2/H2OsolutionwereusedtohydrophilingtheSiO2/Siwafer.TheresultsshowedthatthebesttreatmentsolutionwasH2SO4/H2O2solution,andtheoptimaltreatmenttimewas30minutes.Forthemodificationofmicro-patternedSiO2/Siwafer,thecovalentmethodandhydr
4、ophobizationmethodwerecompared.Itsuggestedthattheformmerissuperiortothelatterinproteinimmobilization.Mercaptoundecanoicacid(MUA)wasusedinthemodificationofthemicro-patternedAu/SiO2wafer.TheresultsofthecontactanglesconfirmedthatMUAhadconnectedontothesurfaceofthemicro-patternedAu/SiO2wafer,whichcould
5、conjugatewithbiomoleculesfurthermore.2.Themicro-patternedSiO2/Siwaferandmicro-patternedAu/SiO2waferwereusedinthedetectionofprotein.Forthesetwokindsofmicro-patternedsiliconwafers,itindicatedthattheoptimalconcentrationfortheimmobilizationofhumanIgGwas100mg/L.Inaddition,averystrongfluorescencesignalw
6、asfoundinthepatternedareaandaverylowfluorescencesignalwasfoundinthenon-patternededarea,whichillustratedthatthetwokindsofmicro-patternedsiliconwafercouldbeusedasthesubstratesofproteinbiochips.Surfaceelementsintwokindsofmicro-patternedsiliconwaferwereanalysedbyX-rayphotoelectronspectroscopy(XPS).The
