2、liestorigintaxainangiosperms.Ithasalonghistoryofcultivationanddiversecultivarswithedible,medicinal,ornamentalvalues.However,classificationandgeneticrelationshiponmanycultivarsremainunclear.Theapproachonlybasedonthetraditionalmorphologyisdifficulttoaccuratelyidentifyacultiva
3、randitsrelationshipwithothers.Inaddition,somewildlotusresourcesarebecomingendangeredTherefore,scientificevaluationandprotectivemeasuresshouldbeimmediatelytakenonlotusgermplasm.Althoughsomemolecularmarkershavebeenrecentlyusedtoinvestigategeneticdiversityandpopulationrelation
4、shipinNelumbo,theapplicationofEST-SSRmarkersandtagnumberareyetlimited.SSRmarkersweredevelopedfromflower-budtranscriptomesequencesofNelumbonuciferainthisstudy,and80polymorphicEST–SSRmarkerstoevaluatethegeneticdiversityofthe139lotusaccessions.Themainresultsareasfollows:1.Inth
5、isstudy,68,593unigeneswereassembledfromamixedflower-budcDNApool,whichwasderivedfromthreeaccessionsofN.nuciferatodevelopSSRmarkers.Atotalof6086SSRlociwereidentifiedanddistributedin5482unigenes,accountingfor8.0%ofthetranscriptomesequences,withanaverageofoneSSRlocusper5.7kbDNA
6、.ThedistributionofSSRlocifrequencywas8.9%.Di-andtri-nucleotiderepeatmotifswerethemajorabundanttypesamong5226SSRloci(removingmononucleotiderepeats)identifiedwithafrequencyof65.2%and31.7%,respectively.Moreover,theAG/CT(57.9%)andGAA/AGA/AAG(13.3%)motifswerepredominantforthedi-
