1、摘要近来大豆分离蛋白(SPI)作为一种新材料资源受到广泛关注。本工作采用乙酸酐和丁二酸酐对SPl进行酰化和羧基化处理制备了乙酰化和羧基丙酰化大豆分离蛋白材料。采用电导滴定法和茚三酮比色法比较测定了酰化度,采用傅立叶红外光谱对其进行了结构表征,并进一步测定了材料的等电点、相对粘度、溶解性、溶液电导率、吸水性、交联凝胶等特性。研究结果表明,酰化度随着反应时间的增加、酸酐的增加而显著增加。酰化度和介质的pH值也有很大的关系,在12附近酰化效果最好。乙酰化SPl的溶解性变化不大;而羧基丙酰化SPI在中性和碱性介质中的溶解性
2、有了很大的提高,在酸性条件下却有所降低。随着乙酰化度的提高,乙酰化sPI等电点降低;而羧基丙酰化SPl的等电点下降幅度较小。乙酰化SPI和羧基丙酰化SPl的相对粘度升高。乙酰化SPI的持水性提高。采用戊二醛对羧基丙酰化SPI进行交联后可得到溶胀度较高的水凝胶。茚三酮比色法测出的酰化度比电导滴定法测出的要高一些,但差别不大于5%:茚三酮比色法和电导滴定法还可以用来测定SPI的分子量,二者测出的SPI的数均分子量的差别不大于2%。关键词:乙酰化大豆分离蛋白;羧基丙酰化大豆分离蛋白;大豆分离蛋白;乙酸酐;丁二酸酐;酰化;
