1、三峡大学硕士学位论文AbstractObjectiveToinvestgatetheeffectsandthemechanismoftotalsaponinsfromPanaxjaponicusC.A.MeyonthebrainaginginducedbyD-galactoseinrats.MethodsWistarratswererandomlydividedintonormalcontrolgroup,lowdosegroup(50mg/kg),middledosegroup(100mg/kg),highdosegroup

2、(200mg/kg)oftotalsaponinsofPanaxjaponicusC.A.Mey.andVEgroup(200mg/kg).Inthemodelgroupanddrug-treatedgroups,D-gal(400mg/kg)wasgiventoratsfor8weeksbysubcutaneousinjection.Morriswatermazewasappliedtodetecttheabilityoflearningandmemoryuntilthe7thweek.After8weeks,thebloo

3、dandbraintissuewereusedinthedictionofindications:thebrainweightindex,theactivityofsuperoxidedismutase(SOD),glutathioneperoxidase(GSH-Px)andtotalantioxidantcapacity(T-AOC),thecontentofmalondialdehyde(MDA)andlipofuscin,thepathomorphismchangesafterHEstainingobservation

4、andtheultrastructurechangesofthehippocampus.Andthen,strsptomycin-biotin-peroxidase(SABC)immunohistochemicalmethodwasappliedtoobservetheproteinexpressionofNrf2.Western-blotmethodwasusedtoanalyizethetheproteinexpressionofNrf2inthenuclesandtheBcl2/Baxinthecells.Real-ti

5、mepolymerasechainreactionwasappliedtodetectthemRNAexpressionsofhemeoxygenasel(HO-1),NAD(P)Hquinineoxidoreductase1(NQO1)andglutamatecysteineligasecatalytic(GCLC).ResultsComparedwiththemodelgroup,thetotalsaponinsromPanaxjaponicusC.A.Mey-treatedgroupsandVE-treatedgroup

6、improvedtheabilityoflearningandmemory:theescapelatencyandthetimeinthetargeted-quadrantwereshorternandtheplatformnumberwasincreased(P<0.05andP<0.01,respectively).Thebrainweightindexwasobviouslyincreasedinthedrug-treatedgroups(P<0.05andP<0.01,respectively).Thedrug-tre

7、atergroupsincreasetheactivityofSOD,GSH-PxandT-AOC,cutdownthecontentofMDAandlipofuscin(P<0.05andP<0.01,respectively).FromHEstaining,thedrug-treatedgroupsobviouslyimprovedthehippocampusnevercelshape,thecellnuclespyknosisandthecellarrangementinneat.Also,thedrug-treated
