1、东南大学硕士学位论文骨髓间充质干细胞移植对兔退变椎间盘髓核细胞衰老影响的体内研究姓名:黄勇申请学位级别:硕士专业:临床医学指导教师:吴小涛2012-05-25中文摘受中文摘要骨髓间充质干细胞移植对兔退变椎间盘髓核细胞衰老影响的体内研究东南大学医学院临床医学院目的:探讨骨髓问充质干细胞(BMSCs)移植对兔退变椎间盘中髓核细胞衰老信号途径的关键因素P16、P53表达的影响,以及对退变椎问盘的修复效应。方法:30只新西兰大白兔的L3/L4、L4/L5、L5/L6椎间盘经髓核抽吸法建立椎问盘退变模型。造模2周后各兔进行BMSCs移植术并分为4组:A组(J下常对照组)为
2、L2/L3,仅行椎间盘暴露;B组(退变对照组)为L3/L4,用微量注射器穿刺髓核一次;C组(透明质酸钠移植对照组)为L4/L5,髓核组织注入透明质酸钠;D组(BMSCs一透明质酸钠移植组)为L5/L6,髓核组织注入BMSCs一透明质酸钠混合溶液。30只兔子分三批,分别于移植术后第2、6、10周三个时间点进行椎I’日J盘MRI扫描并检测T2WI信号强度,HE染色观察椎间盘退变程度,逆转录PCR(RT—PCR)检测髓核组织中P16和P53的mRNA转录水平,Westernblot检测P16、P53的蛋白表达量与II型胶原含量。结果:移植术后第2、6周,D组与C组的T
