1、徐州医学院硕士学位论文葡萄糖受体靶向Gd--DTPA脂质体对肾癌裸鼠模型的MR成像研究姓名:陈超申请学位级别:硕士专业:影像医学与核医学指导教师:徐凯2012-03徐州医学院硕士学位论文葡萄糖受体靶向Gd—DTPA脂质体对肾癌裸鼠模型的MR成像研究中文摘要目的研究葡萄糖受体靶向性Gd—DTPA脂质体进行肾癌裸鼠模型MR成像的可行性,评价其肿瘤特异性。方法超声分散法制备葡萄糖受体靶向Gd—DTPA脂质体用以成像。将人肾透j!jj细胞癌(Kerr-3)细胞注射到裸鼠皮下,构建肾癌裸鼠移植瘤模型。将16只荷瘤裸鼠随机分为实验组和对照组,每组各8只,前者注射葡萄糖受体靶向Gd—DTPA脂质体
2、,后者注射含相同Gd离子浓度的Gd.DTPA,两者均经尾静脉注射。先行T1WI平扫,注射对比剂后5rain、O.5h、lh、2h、3h、4h及5h重复扫描T1WI,扫描结束后测量各时问点瘤体信号强度,绘制瘤体信号强度.时间曲线,以右前肢肌肉组织信号变化作为参照,计算对比噪声L匕(contrasttonoiseratio,.CNR),并进行统计学分析。实验结束后取实验组裸鼠的肝脏、肾脏,做HE病理切片,观察靶向对比剂对裸鼠肝、肾是否有损害。结果实验组注射葡萄糖受体靶向性Gd.DTPA脂质体后5min裸鼠瘤体信号强度开始升高,于3h达到峰值,且5h时强化仍较明显,与平扫比较差别有统计学意
